    Scientific principles

    The total bacterial count detection reagent uses enzyme substrate technology to detect the total bacterial count in water. The reagent contains a variety of unique enzyme substrates, each designed for different bacterial enzymes. When different enzyme substrates are decomposed by enzymes released by different bacteria, they release fluorescent groups. By observing the number of fluorescent cells under the ultraviolet lamp with the wavelength of 365 nm or 366 nm, the total value of colonies can be obtained by looking up the table.

    Science

    Add water sample containing Enterococcus bacteria, culture the target bacteria in Mug medium at  41°C  土  0.5°C,  and  the  specific  切 ological enzymes produced by Enterococcus bacteria(3 -0 -gluco  sidase  can decompose the

    fluorescent substrate mug in the mug medium to produce(3 -D-glucoside ((3 -0 -glucoside) and the

    characteristic fluorescent product 4-methyl­ umbelliferone. Observe the fluorescence in the 366nm UV lamp, count through the quantitative detection disk, and query the MPN table to calculate the results.

    Working Principle
    In the water samples containing total coliform bacteria, the target bacteria were cultured in the ONPG-MUG medium at 36 土 1 C. The specific enzyme betagalactosidase produced by the total coliform bacteria can decompose the color source substrate of the ONPG-MUG medium, which makes the culture medium yellow; meanwhile, Escherichia coli produces a specific beta-glucuronase to decompose the fluorescent substrate MUG in ONPG-MUG medium and produce characteristic fluorescence. The same principle, the heat tolerance coliform group (fecal coliform group) will decompose the color source substrate ONPG in the ONPG-MUG medium at
    44.5 土 0 . 5 °C, making the medium yellow

    The 100ml sterile sampling bottle / quantitative bottle produced by Lifecosm Biotech Limited. is mainly used for the determination of water samples of coliform bacteria by enzyme substrate method. 100ml sterile sampling bottle / quantitative bottle is a product with 51-hole or 97-hole quantitative detection plate,  Lifecosm enzyme substrate reagent and program controlled quantitative sealer. According to the instructions, 100ml water samples were accurately measured with 100ml aseptic sampling bottle / quantitative bottle. The reagents were dissolved in the quantitative detection plate / quantitative hole plate, then seal plate with a program-controlled quantitative sealing machine and culture it about 24h, then count the positive cells. Check the MPN table to calculate.

It is used together with the enzyme substrate detection reagent to accurately determine the MPN value of coliform in 100ml water samples. According to the instructions of  the enzyme substrate reagent, the reagent and the water sample are dissolved, and then poured into the detection plate,  and  then  cultivated after sealed with  sealing machine, the positive pole are counted, then calculate the MPN value in the water sample according to the MPN table
